An experimental model of superoxide dismutase a-induced sarcoidosis-like immune inflammation

The effect of artificially synthesized superoxide dismutase A on the morphogenesis of immune granulomatous inflammation in rats was studied with an assessment of the distribution of subpopulations of antigen-presenting and lymphoid cells in granulomatous infiltrate and determination of immunohistochemical expression of some «targeted» genes/signaling path- ways (PD-L1, PPAR-γ, NF-kB). Descriptive histological and immunohistochemical analysis was performed using CD1a, CD3, CD68 (SCARD1), CD80, CD83, CD163, TBX21, GATA3, Foxp3, IL-17, PD-L1, PPAR-γ and NF-kB markers in the obtained rat tissue samples. In all cases, characteristic morphological signs of sarcoidosis-like granulomatous inflammation with a predominance of the T-cell link, a shift towards type 1 T helper cells against the background of an insignificant number of regulatory T cells were revealed. A significant proportion of monocyte-macrophage cells is determined in the granulomatous infiltrate (mainly due to the M2 subpopulation of macrophages). Features of PD-L1 expression against the background of immune inflammation have been revealed.
