Immunologic factors in tissues of cutaneous melanoma depending on its thickness

The aim was to investigate some characteristics of lymphocytes and cytokines in tissues of tumors and peritumoral area of cutaneous melanoma with various Breslow thicknesses. Fragments of tumor and peritumoral area tissues, as well as the blood of 56 patients with melanoma, were studied. Subsets of lymphocytes in the blood, homogenates of tumor tissues and peritumoral area were studied by the flow cytometry; tissue cytokine levels were determined by ELISA. Increasing melanoma thickness was accompanied by lower levels of CD3+CD4+ lymphocytes, IL-1RА and TNF-α and elevating T-regs and NК-cells, IL-6, IL-8 and partly IL-10 providing immunosuppressive and pro-oncogenic activity of melanoma microenvironment, especially in T3 and T4 tumors. Characteristics of immunologic microenvironment of cutaneous melanoma were specified, and differences in local immunity were demonstrated in tumors depending on the Breslow thickness.
