Expression of transcription factors in tissues of oral mucosa cancer

In the study the expression of HIF-1α and nuclear transcription factors of the NF-kB family, p50 and p65 in tumor tissue of patients with oral mucosa cancer (OMC) were examined. Levels of hypoxia inducible factor-1α (HIF-1α) and nuclear transcription factors of the NF-kB family (p50 and p65) were studied by ELISA in tumor tissue of 41 OMC (T1- 4N0-2M0-1) patients. Histologically unchanged tissue of oral mucosa served as the control. Tumor tissue showed increased levels of HIF-1α – by 2.2 times (p<0.05), NF-kB p50 – by 3 times (p<0.05) and NF-kB p65 – by 6 times (p<0.05), in comparison with the histologically unchanged tissue. Correlation between HIF-1α and NF-kB p50 in tumor tissue was the strongest in patients with poorly differentiated G3 tumors (R=0.56, р<0.0001) and in patients with lymph node lesions (R=0.53, р=0.0001). Correlation between HIF-1α and NF-kB p65 was not so strong.

In conclusions, tumor tissue of oral mucosa cancer showed statistically significant positive correlation between the activation of HIF-1α and NF-kB р50 and NF-kB р65, which was connected with the tumor grade.
