Change of cognition in epileptic children at benzonal treatment depending on individual specificities of its pharmacokinetics

We surveyed children with epilepsy and those receiving antiepileptic drugs: benzonal and valproates. The concentrations of benzonal and its main metabolite – phenobarbital, and valproates in blood plasma were studied. By the results of therapeutic drug monitoring the children treated with benzonal, can be divided into three groups. The children with the concentration of benzonal above 2 mcg/ml and phenobarbital – 10–30 mcg/ml in the blood were referred to the first group. The second group were the patients who have had a minimum benzonal concentration (less than 2 mcg/ml), and the level of phenobarbital was also 10–30 mcg/ml. The third group were the children with low benzonal and high plasma pentobarbital (35–40 mcg/ml). After 1 year of treatment the patients with high levels of phenobarbital in the blood showed a significant reduction of psychophysiological indicators of attention, memory and thinking.
