The atomic force microscopy of the hepatocyte at different degree of the bilirubinemiya

For the first time, depending on the severity of bilirubinemia, the state of the cytoskeleton and the hepatocyte membrane was studied by atomic force microscopy. To study the hepatocyte cytoskeleton, an experiment was performed on 38 minipigs. Atomic force microscopy of hepatocyte was performed on an atomic force microscope IntegraAura (NT-MTD, Russia). As the severity of mechanical jaundice increased, hepatocyte lost the clarity of membrane imaging, the cell nucleus became convex, its segments merged, in animals with the severity of class C mechanical jaundice, the nucleus filled almost the entire cell volume, losing the clarity of the boundaries. Intracellular pressure and hepatocyte volume increased significantly in animals with class B and C mechanical jaundice severity. According to the experimental study, with bilirubinemia increased in animals, the volume of the hepatocyte and its intracellular pressure also increased, the structure of the membrane and the cytoskeleton of the hepatocyte changed.
