The influence of induced diabetes mellitus in rats on IGF -axis during growth of Gerena carcinoma

The IGF system through paracrine/autocrine mechanisms regulates the growth, proliferation, survival and differentiation of normal and malignant cells. The aim is to study the effect of such comorbid pathology, as diabetes mellitus on the content of IGF and proteins binding them in the blood in female outbred white rats with transplanted Guerin’s carcinoma. White outbred rats of both sexes (n=64) were divided into groups with induced alloxan diabetes, subcutaneous inoculation of Guerin’s carcinoma, and the combination of these processes. The ELISA method was used to determine the level of IGF1, IGF2, IGFBP1, IGFBP2 in the animals blood. As it was shown in rats of both sexes against the background of diabetes mellitus, there was unidirectional shifts in the content of IGFBP1 and IGFBP2 occurred, while changes in the IGF level turned out to be specific: IGF1 in females increased 1.5 times (p<0.05), and IGF2 decreased 2.9 times, while in males no changes in IGF were detected in the blood, and IGF2 increased by 2.2 times. Transplantation of Guerin’s carcinoma caused unidirectional changes in the studied factors, regardless of the animals sex, which were expressed in decreased level of IGF1, IGFBP1, IGFBP2 in the blood. As the result of the combination of carcinoma growth with endocrine pathology, were noted the changes in the content of IGF and IGFBP, which had sex specificity, that led to a modification of the malignant process course.
